Human serum IgG comprises four subclasses, IgG1 (60-66%), IgG2 (20-30%), IgG3 (less than or equal to 5%) and IgG4.1 Individual subclasses vary with respect to their initial amino acid sequence, their physical and chemical properties and age dependent serum concentrations.2 Antigen recognition, Complement activation, cell surface-receptor binding, blocking activity, and possibly placental transfer are among the biologically important functional differences between the four human IgG subclasses.3 Every subclass has a specific biological function:
response to protein antigen is mainly mediated by IgG1 and IgG3, whereas IgG2 mediates response to polysaccharide antigens. IgG1 and IgG3 can bind to the cells that mediate immune response. IgG4 activates Complement using an alternative route.2 An isolated absence of a certain specific subclass may or may not be associated with disease. IgG subclass deficiency should be looked into especially when conditions/ diseases like 1) recurrent bacterial infections of respiratory and digestive tracts often caused by pyogenic microorganisms, 2) IgA deficiency, 3) absence of immune response following vaccination, 4) allergic or autoimmune diseases are diagnosed.2
This Human IgG pincerTM Assay is a rapid, homogeneous antibody-based fluorescence assay for detecting and quantifying human IgG in hybridoma cell supernatants. The assay uses two human IgG pincers that are anti-human IgG specific antibody conjugated to short complementary single-stranded oligonucleotides. In the presence of human IgG, the two single-stranded oligos associate to form a duplex and FRET (Fluorescence resonance energy transfer) is initiated between the two fluorophores labelled at the end of the two oligonucleotides. The amount of FRET signal is associated with the IgG concentration present in the samples. This assay is simple (one step) and fast (30 min).
