In mice, IgG comprises four subclasses : IgG1, IgG2a, IgG2b and IgG3. Mouse IgG1 plays an important role in recognition of carbohydrate epitopes. IgG2a and IgG2b have the ability to fix Complement and recognize protein antigens. IgG3 is mainly involved in binding to Mast cells [Hussain R et al. (1995) Clin Diagn Lab Immunol. 2(6): 726-732] Individual subclasses vary with respect to their initial amino acid sequence, their physical and chemical properties.
This Mouse IgG pincerTM Assay is a rapid, homogeneous antibody-based fluorescence assay for detecting and quantifying mouse IgG in biological samples such as ascitic fluids and hybridoma cell cultures. The assay uses two mouse IgG pincers that are anti-mouse IgG specific antibody conjugated to short complementary single-stranded oligonucleotides. In the presence of mouse IgG, the two single-stranded oligos associate to form a duplex and FRET (Fluorescence resonance energy transfer) is initiated between the two fluorophores labelled at the end of the two oligonucleotides. The amount of FRET signal is associated with the IgG concentration present in the samples. This assay is simple (one step) and fast (30 min).
